piRNA Array Service

piRNAs are a class of non-coding RNAs of approximately 26-32 nucleotides in length. The piRNA interacts with the PIWI (P-element-induced Wimpy Testis, subfamily of the Argonaute protein) to form a piRNA/PIWI complex, thereby silencing the transposable element to maintain genomic integrity, as well as phylogeny and spermatogenesis. The piRNA is produced by a primary transcript that traverses the entire piRNA cluster and is subsequently processed into mature piRNA. Although piRNA has been found in large numbers in many species, there are many unresolved problems in the field of piRNA that need further study.

Workflow of piRNA array Service

  • RNA isolation (optional)
  • RNA QC
  • The absorbance of RNA at 260 nm, 280 nm, and 230 nm of the spectrophotometer is measured to calculate the concentration and evaluate the purity
  • Denaturing agarose gel electrophoresis is carried out with formaldehyde electrophoresis reagent to detect RNA purity and integrity
  • Provide RNA QC reports

Note: RNA samples used for chip detection must be of high quality, intact, free of RNase contamination (degraded samples cannot be used for labeling and chip detection) and are free of genomic contamination.

  • Target repair with Cy3 tag
  • Array hybridization, cleaning and scanning
  • Data extraction, analysis and summary
  • Experimental reports - including detailed experimental methods and chip experiment data and charts

  • Operating instructions (including RNA quality inspection report)

  • Hierarchical clustering (multiple samples are grouped according to gene expression similarity when multiple sample experiments are performed)

If repeating the experiment (≥3), calculate the CV value, p-value, and do Volcano Plot
