IntegrateRNA has successfully implied CRISPR/Cas9 system into loss-of-function studies of lncRNA. LncRNAs are defined as the noncoding linear transcripts longer than 200 nucleotides, which play important roles in the regulation of transcription and post-transcriptional processes of coding and non-coding RNAs. Moreover, lncRNAs have been found to function as novel types of oncogenes and tumor suppressors during cancer progression through various mechanisms, which endow them with the potential of serving as reliable biomarkers and novel therapeutic targets for cancers. For many lncRNAs with complex transcriptional profile and regulatory regions, the CRISPR/Cas9 mediated ablation of the locus can act as a very specific and powerful tool to study their functions both in vitro and in vivo.

As shown, Cas9 can be directed by a pair of specifically designed sgRNAs targeting the 5' and 3' ends of the lncRNA gene to generate lncRNA fragment deletion. This method is particularly useful for performing CRISPR lncRNA knockout, as simple indel mutations do not facilitate effective knockout of lncRNA. Besides, we also utilize a paired-guide RNA library to achieve whole genome-scale delection of lncRNAs. This is convenient for high-throughput screening of lncRNA knockout phenotypes.
